Precautions and warnings
Hepatotoxicity cases reported in patients treated with Crizotinib capsules have fatal outcomes. Monitor liver function tests, including ALT, AST, and total bilirubin, every 2 weeks during the first 2 months of treatment, then once a month. Withhold, reduce dose, or permanently discontinue Crizotinib for hepatotoxicity.
Severe or fatal interstitial lung disease (ILD)/pneumonitis can occur in patients treated with Crizotinib. Monitor patients for pulmonary symptoms indicative of ILD/pneumonitis and permanently discontinue Crizotinib in patients diagnosed with drug-related ILD/pneumonitis.
Symptomatic bradycardia can occur in patients treated with Crizotinib. Avoid using this drug in combination with other medications known to cause bradycardia. One should regularly monitor heart rate and blood pressure and withhold, reduce dose, or permanently discontinue Crizotinib for bradycardia.
According to its mechanism of action and findings in studies, Crizotinib can cause fetal harm when administered to a pregnant woman. Based on the potential for genotoxicity, advise males with female partners of reproductive potential to use condoms during treatment with Crizotinib and for 3 months following completion of therapy.
Interaction
Administration of Ibrutinib with other drugs will affect the mechanism of Ceritinib. These drugs are beta-blockers, non-dihydropyridine calcium channel blockers, clonidine, and digoxin.
